Cai Lei, frmer vice-president f JD Grup, reveals hw amytrphic lateral sclersis (ALS) diagnsis (诊断) set him n the path t find a cure.

Alchl misuse is the biggest risk factr fr death, disability and ill health amng peple aged 15 t 49 in the UK, respnsible fr 9,641 deaths during 2021. Like ther addictive substances, alchl triggers the release f a feel gd chemical in the brain called dpamine. Hwever, lng-term drinking decreases dpamine release, and peple wh are addicted t alchl dn’t tend t feel pleasure in drinking it any mre. “It seems that they’re drinking mre because they feel a need t maintain a drunk state,” said Prf Kathleen Grant at Oregn Health and Science University.
The idea f the gene therapy was t try t reset this dpamine reward pathway by enhancing the functin f brain cells that synthesize dpamine. Like humans, sme macaque mnkeys are mre likely t drink heavily than thers: if yu begin t ffer them the chice f 5% alchl alngside their regular fd and water, sme mnkeys will eventually almst always chse the alchl. Grant and her clleagues wndered whether resetting their dpamine reward pathways might cntain their desire t drink alchl. T d this, they used a harmless virus t deliver the gene fr a prtein called glial-derived neurtrphic factr (GDNF) t an area f the brain that is invlved in addictin and reward. “We targeted the cell bdies that prduce dpamine with this gene t increase dpamine synthesis, thus supplementing r restring what lng-term drinking has taken away,” Grant said.
The research, which was published in Nature Medicine, fund that the macaques permanently started verexpressing dpamine and decreased their alchl cnsumptin by up t 90%. “We are entering an era f gene therapy fr neurlgical disrders and perhaps mental disrders, and I think this study is very prmising in that directin.” Grant said.
