The risk f develping Parkinsn’s disease is twice as high in men as in wmen, and new research pints t a pssible reasn why: a prtein (蛋白质) in the brain.
PTEN-induced kinase 1 (PINK1) prtein is nt nrmally a threat, and is imprtant in cntrlling cellular energy use in the brain. Hwever, the new research shws that in sme Parkinsn’s cases, the immune system mistakes PINK1 fr an enemy, attacking brain cells that express the prtein.
Accrding t the study, led by a team frm the La Jlla Institute fr Immunlgy in Califrnia, the PINK1-related damage dne by the immune system’s T cells is much mre widespread and aggressive in the brains f men than wmen. “The sex-based differences in T cell respnses were very striking,” says Alessandr Sette, “This immune respnse may be a cmpnent f why we see a sex difference in Parkinsn’s disease.”
Using bld samples frm Parkinsn’s patients, the researchers tested the respnse f the T cells in the bld against a variety f prteins previusly linked t Parkinsn’s—finding PINK1 std ut. In the male Parkinsn’s patients, the team nticed a six-time increase in T cells targeting PINK1-tagged brain cells, cmpared with healthy brains. In the female Parkinsn’s patients, there was nly a 0.7-time increase.
As is always the case with research f this type, nce experts knw mre abut hw a disease gets started and hw it prgresses, that pens up new pprtunities fr finding ways t stp the damage. “We culd pssibly develp treatments t blck these T cells, nw that we knw why the cells are targeting in the brain,” says Cecilia Lindestam Arlehamn.
Further dwn the line, being able t discver these PINK1-sensitive T cells in bld samples culd lead t Parkinsn’s disease being diagnsed at an earlier stage—which again helps with treatment and patient supprt. While we’re waiting t discver a cure fr Parkinsn’s disease, cnstant prgress is being made in understanding the risk factrs invlved in its develpment, and new appraches t dealing with it.
